自噬是什么?

“自噬”一词并不是随机选择的,它的意思是“自我吞噬”。自噬是一种分解代谢过程,会分解身体细胞的成分,以便将它们用于创建新细胞。这是一种称为稳态的回收机制。

 

细胞质是一种“果冻状”物质,位于细胞核之外。当自噬发生时,细胞质和具有独特功能的微小结构(称为细胞器)被从细胞中移除并回收。这个过程至关重要,可以清除不再正常工作的细胞,从而保持身体平衡。有几种疾病(尤其是神经退行性疾病),例如帕金森病,会破坏自噬过程。

 

它是如何工作的? 

当细胞没有获得足够的营养时,自噬就会开始。自噬过程分为四个阶段:

 

1. 碳封存

 

在此步骤中,吞噬泡(双层膜)在细胞质和细胞器周围移动,直到它们完全封闭。然后吞噬泡变成一种称为自噬体的细胞器。

 

2. 融合

 

自噬体不能直接附着于溶酶体。因此它开始与称为内体的结构合并。当自噬体与内体融合时,它被称为两栖体。两性体具有与溶酶体融合的能力。

 

3. 降解

 

两性体与溶酶体融合后,降解开始发生——溶酶体释放水解酶(一种酶),降解先前被自噬体包围的物质。  具有现已降解的细胞材料的结构称为自噬溶酶体或自溶酶体。

 

4. 重复使用

 

在细胞材料完全降解、转化为氨基酸并从自噬溶酶体直接输出到细胞液中后,这些氨基酸现在可以被新细胞重复使用。

 

分解的氨基酸用于 TCA 循环(通常称为柠檬酸循环)。这是一系列化学反应,是细胞呼吸的核心驱动力。 NAD+ 是我们最畅销的补充剂之一,在 TCA 循环内的大多数反应中发挥着重要作用。


自噬的不同类型 

 

自噬可分为三种类似的类型,但具有不同的特征:

 

1. 巨自噬

这是指如上所述的一般自噬过程。

 

2. 微自噬
这个过程还会吞噬并降解不同的细胞结构;然而,它在隔离过程中不利用吞噬细胞。相反,溶酶体吸入细胞内容物并开始吞噬其膜周围的物质。然后内容物被分解成氨基酸,可以回收利用。

 

3. 伴侣介导的自噬
这是一种选择性自噬过程,其中蛋白质被靶向以被降解。所谓的伴侣蛋白有助于可降解蛋白沿着溶酶体膜易位。

 

抗衰老和长寿

自噬是一种压力(压力是细胞饥饿)反应,它可以使细胞恢复活力,并使它们在能量使用方面变得更加保守,并且对损伤更有弹性。研究表明,自噬的激活可以抑制与年龄相关的损坏细胞结构的积累,并大大提高目标细胞的代谢效率。[i]

 

还可以触发自噬来清除故障线粒体,这些线粒体会产生许多有害的 ROS(活性氧),从而导致细胞降解。  这个过程被称为线粒体自噬。[ii

 

诱导自噬也被证明可以延长小鼠的寿命。[iii]

 

自噬的其他好处

自噬不仅具有抗衰老作用,还可能在预防与年龄相关的疾病方面发挥关键作用。自噬可以清除已知会导致神经退行性疾病(例如阿尔茨海默病和帕金森病)发展的有毒蛋白质。

 

许多不再发挥作用的细胞会通过突变和繁殖造成伤害,这构成了每种癌症传播的基础。自噬通过将细胞内损坏的结构分解成氨基酸来防止故障细胞增殖。因此许多医学专业人士认为自噬在癌症的预防和治疗中起着至关重要的作用。身体检测出问题,将其摧毁,并进行“修复工作”,从而提高基因组的稳定性。虽然没有具体的科学证据支持这一假设,但一项研究的结果提供了支持这一理论的进一步迹象。[iv]

诱导自噬的方法
 

除了抗衰老之外,还有如此多令人难以置信的健康益处,您可能会问自己如何触发体内的自噬。如前所述,自噬是一种应激反应。因此,不会严重损害我们身体的轻微压力可以通过激活自噬而有益。大量研究帮助确定了每天可以采取的几种措施来诱导自噬。最为显着地:

3.间歇性禁食和热量限制

4. 充足的睡眠
