在哺乳动物中发现了 7 种 Sirtuins,但研究最多的是 SIRT1,其作用之一是热量限制有助于延长寿命。热量限制 (CR) 是延长线虫、果蝇、小鼠甚至猴子等多种生物体寿命的研究最多且可靠的方法之一。

CR 作为一种延长人类寿命的方法,很难通过科学研究进行研究,这是可以理解的,但最终证明它可以减少一些与年龄相关的疾病的危险因素,这些疾病会缩短人类的寿命。有趣的是,当小鼠的 SIRT1 基因被禁用并且这些小鼠被限制热量饮食时,寿命的延长以及表明抗衰老作用的血液标记物并没有发生,这表明 CR 如何依赖 Sirtuins 来发挥作用。激活延长寿命的机制。

SIRT1 不仅是产生热量限制的抗衰老作用所必需的,CR 本身还可以提高人体内 Sirtuin 的产量和水平。间歇性禁食也是如此。 CR 和间歇性禁食都与较低的胰岛素水平以及较低的 IGF-1(胰岛素样生长因子)水平有关。

Sirtuins 和 NAD+

SIRT1 的作用是去除这些乙酰基,从而保持蛋白质的健康和功能。但如果没有 NAD+ 的存在,SIRT1 就无法发挥作用,NAD+ 是催化去除乙酰基反应所必需的辅酶。
