Can NMN Supplementation Treat Alzheimer’s Disease?

Kan NMN-supplementering behandle Alzheimers sygdom?

Kognitiv tilbagegang er en uheldig del af aldringsprocessen.

Efterhånden som vi bliver ældre, øges vores risiko for Alzheimers sygdom (AD). Den neurodegenerative lidelse påvirker hjernens kognitive og hukommelsesfunktioner – og de nuværende behandlingsmuligheder er begrænsede. I dag anslås det, at AD påvirker 44 millioner mennesker verden over.

Selvom der ikke er nogen kendt kur mod AD, kan tilskud muligvis bremse eller forhindre sygdommens progression. Studier udført på virkningerne af nikotinamidmononukleotid (NMN) tilskud i mus og rotter afslører for eksempel betydeligt behandlingspotentiale.

I dette indlæg vil vi undersøge potentialet af NMN som en behandling for kognitiv tilbagegang og aldersrelaterede sygdomme som AD. Vi vil diskutere, hvad NMN er, undersøge, hvordan det virker, og udforske begrænsningerne i den nuværende forskning om, hvordan det muligvis kan forbedre symptomerne på AD.

 

Baggrund om Alzheimers sygdom 

  

AD er en progressiv hjernelidelse, der resulterer i neuronal skade og nedsat kognitiv funktion. Sygdommen er den mest almindelige årsag til demens – den påvirker patienternes hukommelse, humør og adfærd.

Alzheimers sygdom begynder generelt langsomt og bliver værre over tid. Gradvist kan det forstyrre en persons daglige liv. Tidlige symptomer på AD inkluderer:

 

- Glemsomhed

- Desorientering

Sværhedsgrad ved udførelse af opgaver

 

Efterhånden som symptomerne udvikler sig, kan AD-patienter også opleve:

 

Humør- og adfærdsændringer

Sprogproblemer

Nedsat dømmekraft

 

Selvom der ikke findes nogen kur mod AD, kan visse medicin, kosttilskud og andre behandlinger hjælpe patienter med at håndtere deres symptomer. Forskning er i gang – og studier indikerer, at NMN-tilskud kan spille en rolle i at forhindre neuronal skade og bremse udviklingen af AD.

 

Hvad er NMN?

En påmindelse om, at når en person bliver ældre, bliver de mere modtagelige for neuronale skader og kognitiv tilbagegang.

NMN kan muligvis hjælpe med at modvirke dette. Studier viser, at det naturligt forekommende molekyle har neurobeskyttende effekter.

Produceret af kroppen, er NMN en forløber for nicotinamid-adenin-dinukleotid (NAD+): et coenzym, der spiller en vigtig rolle i DNA-reparation, energimetabolisme og cellulær signalering.

NAD+ findes i alle kroppens celler. Uden det ville cellerne ikke kunne overleve.

 

Dette er ikke et problem hos unge mennesker. Tidligt i livet har vi al den NAD+, vi har brug for. Men med tiden falder niveauet af NAD+ i kroppen betydeligt. Dette fører til cellulær skade, energimangel og andre helbredstilstande. Eksperter har fundet ud af, at en mangel på NAD+ ikke kun bidrager til aldersrelateret tilbagegang, men også til opståen af helbredstilstande som AD.

Dette er, hvor tilskud kan komme ind i billedet. Ved at supplere med NMN mener forskere, at kroppen kan øge sin produktion af NAD+ og i sidste ende bremse aldringsprocessen.

 

NMN som en potentiel behandling for AD

NMN-supplementering har vist sig at forhindre kognitiv tilbagegang hos ældre mus og rotter med AD. Selvom forskningen er noget begrænset, kan resultaterne meget vel overføres til mennesker.

Eksisterende dyreforsøg viser stor lovende potentiale. For eksempel undersøgte en nylig undersøgelse offentliggjort i Innovations in Aging effekterne af NMN-supplementering på kognitiv funktion hos ældre mus (1). Undersøgelsen fokuserede på 24 måneder gamle mus (svarende til 70-årige mennesker) med AD. Musene blev behandlet med NMN over to uger.

Forskerne fandt ud af, at justering af cerebral blodgennemstrømning (CBF) via neurovaskulær kobling (NVC) – en kritisk mekanisme, der forsyner hjernen med energisubstrater fra blodet – kunne hjælpe med at opretholde en sund hjernefunktion hos musene.

Nedsat NVC fører derimod til kognitiv tilbagegang. Og der er beviser for, at et fald i NAD+ over tid kan forværre problemet. Faktisk så forskere, at de ældre mus' NVC-responser var nedsatte – men de fandt, at NMN-supplementering kunne genoprette deres NVC-responser.

Undersøgelsen fastslog til sidst, at et fald i NAD+-forsyningen bidrog til aldersrelateret cerebral dysfunktion hos musene, hvilket forstærkede deres kognitive tilbagegang (1). Med NMN-supplementering forbedrede musene dog deres koordination og rumlige arbejdshukommelse. Med andre ord begyndte de at overvinde nogle af deres AD-symptomer.

Dette styrker NMN som en potentiel AD-behandling eller neurobeskyttende løsning for mennesker også.

 

Flere beviser for, at NMN kan forbedre kognition

En anden undersøgelse offentliggjort i Brain Research udførte lignende forskning – denne gang på rotter med AD (2). Forskerne fandt, at NMN-supplementering kunne genoprette kognitiv funktion og mindske amyloid beta (Aβ) ophobning i de kognitivt svækkede rotter.

Aβ, det er værd at bemærke, er et protein, der ophobes i hjernen. Aβ-toksicitet er forbundet med AD – og undersøgelsen fandt, at NMN-supplementering reducerede Aβ-ophobning. Det hjalp også med at forbedre neuronoverlevelse, øge energimetabolismen og reducere ophobningen af reaktive iltarter (ROS).

ROS er et ustabilt molekyle, der spiller en rolle i kognitiv svækkelse. De øgede niveauer af NAD+ i rotternes hjerner fremmede sund kognition ved at aktivere et protein kaldet SIRT1 og producere antioxidative enzymer.

Resultatet var reduceret neuronal skade og øget kognitiv funktion hos rotterne (2). Resultaterne tyder på, at NMN-supplementering kan hjælpe rotter med AD med at forhindre kognitiv skade og genoprette deres neuronale sundhed. Dette viser, at NMN-supplementering kan tjene som en tilsvarende behandlingsmulighed for menneskelige patienter med AD og andre neurodegenerative sygdomme.

 

Konklusion

NMN-supplementering har vist sig at tilbyde neurobeskyttende effekter hos ældre mus og rotter med AD. De kognitive forbedrende effekter af NMN-supplementering er resultatet af aktiveret SIRT1 og fremme af antioxidative veje. Disse fordele vil sandsynligvis overføres fra dyremodeller til menneskelige modeller – hvilket hjælper folk med at forhindre neuronal skade, når de ældes.

I de senere år har NMN-tilskud været i fokus som en måde at forebygge eller bremse aldringsprocessen. Selvom der er behov for mere forskning på mennesker, tilbyder molekylet store løfter. NMN kan også forbedre synet, afbøde høretab og forbedre den generelle fysiologiske sundhed.
