Hvad er Ergothioneine (ERG)?

Helblod, urin og spytmetabolomik
Metabolomics er studiet af små molekyler (metabolitter) i biologiske prøver, såsom blod, urin og spyt, for at forstå fysiologiske og patologiske tilstande. Forskere har brugt metabolomics til at undersøge rollen af ERG og andre forbindelser i aldersrelaterede sygdomme.

Sult, ERG og aldring
Undersøgelser har vist, at kalorierestriktion (CR) eller periodisk faste (IF) kan forlænge levetiden og reducere oxidativt stress i forskellige organismer. Disse fordele tilskrives delvist aktiveringen af specifikke transkriptionsfaktorer, der opregulerer antioxidantgener. Spaltningsgæren, S. pombe, fungerer som en fremragende modelorganisme til at studere virkningerne af sult på stofskiftet, da den deler mange ligheder med menneskelige celler.

Forskere observerede, at både glukose- og kvælstofsult førte til betydelige stigninger af ERG i S. pombe. Ligeledes fandt forskere i en lille undersøgelse med fire unge, ikke-overvægtige menneskelige frivillige, der fastede i 58 timer, at niveauerne af ERG og andre antioxidanter i deres blod steg. Disse fund tyder på, at stigningen i ERG-niveauer kan være en adaptiv respons på faste-stress hos både gær og mennesker, og potentielt spille en beskyttende rolle i aldringsrelaterede processer.


ERG i skrøbelighed, demens og sarkopeni
Skrøbelighed, demens og sarkopeni er almindelige aldersrelaterede sygdomme, der deler kliniske træk, såsom øget afhængighed af livsstøtte. Forskere brugte helblodsmetabolomik til at studere disse sygdomme og opdagede, at ERG-niveauerne faldt markant ved skrøbelighed og demens, men ikke ved sarkopeni (en tilstand karakteriseret ved tab af muskelmasse og funktion). Desuden faldt ERG-relaterede forbindelser, S-methyl-ERG og hercynin, også ved skrøbelighed og demens. Denne opdagelse antyder, at indtagelsen eller metabolismen af ERG kan være påvirket i disse tilstande. Faldet i ERG, en potent antioxidant, kunne bidrage til udviklingen af skrøbelighed, demens og andre aldersrelaterede hændelser, da oxidativ skade er kendt for at fremskynde disse lidelser.

Forskere analyserede også urin- og spytmetabolomik for at opnå en mere omfattende forståelse af aldersrelaterede sygdomme. De fandt, at ERG-niveauerne i disse biofluider ikke viste betydelige aldersrelaterede forskelle, hvilket indikerer, at yderligere forskning er nødvendig for at forstå ERG's rolle i disse tilstande.
