Ergothioneine: A Promising Biomarker Linking Health-Conscious Food Patterns to Reduced Cardiometabolic Disease Risk and Mortality

Ergothionein: En lovende biomarkør, der forbinder sundhedsbevidste madmønstre med reduceret risiko for kardiometaboliske sygdomme og dødelighed

Denne artikel diskuterer en befolkningsbaseret prospektiv undersøgelse, der havde til formål at identificere plasmametabolitter forbundet med et sundhedsbevidst kostmønster (HCFP) og en lavere risiko for kardiometabolisk sygelighed og dødelighed under en langvarig opfølgning. Undersøgelsen fandt, at øgede niveauer af aminosyren ergothioneine var stærkt og uafhængigt forbundet med både HCFP og en lavere risiko for fremtidig koronararteriesygdom (CAD), kardiovaskulær og al-dødelighed. Disse fund antyder nye veje, der forbinder kost med kardiometabolisk sundhed.

 

Flere metabolitter forbundet med HCFP er tidligere blevet korreleret med selvrapporteret indtagelse af specifikke fødevaregrupper eller -varer. Ergothioneine findes i mange kostkilder og har særligt høje niveauer i svampe, tempeh og hvidløg. Det er tidligere blevet forbundet med et højere indtag af grøntsager, fisk og skaldyr samt et lavere indtag af faste fedtstoffer og tilsat sukker, og det er også blevet forbundet med sunde madmønstre. Dette er i overensstemmelse med undersøgelsens resultater vedrørende sammenhængen mellem ergothioneine, indtagelse af grøntsager, fisk og skaldyr og HCFP.

 

Proline betain, også kendt som stachydrin, og methylprolin er begge kendte biomarkører for citrusfrugtindtag, hvilket kunne forklare deres association med frugtindtag i denne undersøgelse. Acetylornithin er blevet forbundet med et højere indtag af grøntsager, hvilket også blev bekræftet i denne undersøgelse. Pantothenat, også kendt som vitamin B5, er bredt fordelt i alle fødevaregrupper. I modsætning hertil har urobilin, som viste en negativ association med HCFP i denne undersøgelse, ikke tidligere været forbundet med noget kostindtag. Korrelationen mellem metabolitniveauer og fødevaregrupper var beskeden, men korrelationskoefficienterne mellem ergothionein og fødevaregrupper var lignende tidligere rapporterede værdier. På trods af den omfattende kostprøvetagningsmetode anvendt i Malmö Diet and Cancer (MDC) study, er måleinaktigheder sandsynligvis med til at svække de observerede korrelationer.

 

Ergothioneine havde den stærkeste association med HCFP og de mest tydelige beskyttende associationer med kardiometabolisk morbiditet og mortalitet, uafhængigt af traditionelle risikofaktorer. Resultaterne antyder ergothioneine som en biomarkør for både sundt kostindtag og lav risiko for fremtidig kardiometabolisk sygdom. Ergothioneine har vist sig at beskytte gnavere mod iskæmisk reperfusionsskade og er også blevet foreslået som en antioxidant med potentielt gavnlige effekter på menneskekroppen. Ergothioneine adskiller sig fra andre foreslåede antioxidanter ved at have en specifik transportør, der er blevet foreslået at være opreguleret i områder med inflammation, hvilket giver ergothioneine potentiale for en mere kontrolleret antioxidantfunktion. At have højere niveauer af ergothioneine kunne beskytte mod oxidativt stress på en reaktionær måde, hvilket menes at være en vigtig faktor i patogenesen af kardiovaskulær sygdom (CVD) og kunne forklare undersøgelsens fund."

 

En nylig undersøgelse udført på raske mennesker viste, at oral administration af ergothioneine øgede niveauerne af cirkulerende ergothioneine og nedsatte niveauerne af nogle biomarkører for oxidativ skade. Interventionsforsøg med randomiserede behandlingsregimedesign er nødvendige for at undersøge, om denne potentielle antioxidanteffekt kan mindske risikoen for kardiometabolisk sygdom.

Den positive korrelation mellem ergothioneine og alkoholindtag er tidligere blevet demonstreret, en sammenhæng der enten kan forklares ved, at ergothioneine er til stede i alkoholiske drikkevarer, eller ved at alkohol ændrer absorptions effektiviteten af ergothioneine, der er til stede i andre kostkilder.

 

Afslutningsvis fandt denne undersøgelse, at højere niveauer af ergothionein var forbundet med en lavere risiko for kardiometabolisk sygdom og dødelighed, hvilket antyder, at en specifik sund kost potentielt kunne påvirke disse resultater ved at påvirke specifikke metaboliske veje og mekanismer. Den stærke og uafhængige association af ergothionein med både HCFP og en lavere risiko for fremtidig CAD, kardiovaskulær og al-dødelighed fremhæver vigtigheden af at forstå de molekylære begivenheder, der resulterer fra kostindtag og deres forhold til sygdoms- og sundhedsresultater. Denne viden ville lette fremtidige interventionsstudier ved at identificere kost-modificerbare metaboliske veje og sygdomsmekanismer, hvilket muliggør design af mere effektive kostinterventioner for at forbedre kardiometabolisk sundhed.
