The newest findings on NAD+, the Circadian Rhythm, and Anti-aging

De nyeste fund om NAD+, den cirkadiske rytme og anti-aging

Mens rollen af den cirkadiske rytme og de anti-aging fordele ved NAD+ allerede er velkendte, giver en ny banebrydende undersøgelse, der blev offentliggjort for blot en uge siden, nye indsigter i:[i]

 

- I hvilket omfang NAD+ kan ændre genekspressionen af døgnrytmen.
- Hvordan NAD+, med hjælp fra SIRT1, stabiliserer BMAL1-aktivitet ved at undertrykke PER2, og hvordan dette fremmer cirkadian transkription.
- Hvordan NAD+ tilskud ændrer døgnrytmen, genopretter undertrykt BMAL1-binding, cellulære oscillationer, respirationsrytmer og aktivitetsrytmer tilbage til ungdommelige niveauer.

 

Hvad er den cirkadiske rytme?

Den cirkadiske rytme beskrives nogle gange også som kroppens indre ur, der regulerer søvnighed og vågenhed i løbet af dagen. Den cirkadiske rytme styres af et område i hjernen, der er meget følsomt over for lysændringer. Dette er grunden til, at vi er mest opmærksomme, når solen skinner, og bliver trætte, når det bliver mørkt.

Hvad er NAD+?

NAD+ er et essentielt molekyle, der kan findes overalt i din krop. Det er en nøglekomponent i omkring 500 forskellige enzymatiske reaktioner, der sker i vores kroppe [ii]. NAD+ kan suppleres gennem forløbere, såsom NMN (Nicotinamidmononukleotid) og NR (Nicotinamidribosid) [iii].

 

 

Hvad vides der i øjeblikket om døgnrytmen og NAD+?

Efterhånden som vi bliver ældre, begynder vores døgnrytme at svækkes – vi føler os mindre vågne, når vi udsættes for sollys, og mindre søvnige, når det er mørkt. Grundlæggende bliver vores krops indre ur dæmpet [i]. Sammen med nedgangen i døgnrytmen falder NAD+-niveauerne også, når vi bliver ældre, så naturligvis har forskere været nysgerrige på, om der er en tovejs korrelation mellem NAD+-niveauer og døgnrytmen.

 

In vivo- og in vitro-studier har vist, at NMN-supplementering (som øger NAD+-niveauerne) forlænger levetiden for organismer som mus [ii] og orme samt mikroorganismer som gær [iii]. Derudover har NMN-supplementering vist sig at beskytte mod fysisk tilbagegang, der tilskrives aldring, såsom muskelregenerering, et fald i fysisk formåen, mitokondriel dysfunktion, svækkelse af synet, insulinresistens, arteriel dysfunktion og mere [iv].


En undersøgelse offentliggjort den 4. maj 2020 giver os nye indsigter i, hvordan NAD+ påvirker den cirkadiske rytme.

 

Denne in vivo-undersøgelse undersøgte NR (nicotinamid ribosid) supplementering (400 mg/kg/dag) hos mus over en periode på fire måneder og sammenlignede det med en kontrolgruppe af mus, som i stedet blev fodret med almindeligt vand. NR er en anden NAD+ forløber, ligesom den førnævnte NMN. Efter fire måneder blev musenes gener undersøgt; deres genekspression ændrede sig drastisk. Omkring 50% af musenes gener viste en betydelig ændring i ekspression. Nogle gener:

 

1. Viste et tab i døgnrytmeoscillation
2. Viste en stigning i døgnrytmeoscillation
3. Viste en ændring i udtrykket af døgnrytme
4. Var upåvirkede (omkring 50%)

 

Selvom disse var bemærkelsesværdige fund, opstod der et vigtigere spørgsmål. Hvordan opnår NAD+ disse ændringer?
 

Undersøgelsen begyndte at undersøge rollen af BMAL1, som er et protein, der er involveret i transskriptionen af forskellige gener, der påvirker de cirkadiske urmekanismer i alle pattedyr, inklusive mennesker. Mus blev opdelt i to grupper. Den ene havde normale NAD+ og BMAL1 niveauer, mens den anden gruppe bestod af mus, der var mangelfulde i BMAL1. Begge grupper blev injiceret med 500 mg/kg NMN (NAD+ forløber), og DNA-prøver blev indsamlet fire timer senere. Efter at have undersøgt BMAL1-bindingerne i prøverne, blev det konkluderet, at NAD+ øger cirkadisk transskription ved at stabilisere BMAL1.

 

Men for at NAD+ effektivt kan stabilisere BMAL1-bindinger, kræves tilstedeværelsen af SIRT1. SIRT1 er en sirtuin, en gruppe af NAD+ afhængige proteiner. SIRT1 er også en proteindeacetylase. Proteindeacetylaser er enzymer, der fjerner acetylgrupper fra lysin (en almindelig aminosyre/protein). Ved at se på celler, der ikke indeholder SIRT1, identificerede de øgede niveauer af PER2 i kernen af disse celler. PER2 er et protein, der er kendt for at undertrykke BMAL1-aktivitet.

 

Baseret på disse fund kom de til den konklusion, at: SIRT1 fjerner acetylgruppen fra PER2-proteiner, hvilket ændrer PER2, og dermed reducerer dets effektivitet i at undertrykke BMAL1-aktivitet. BMAL1-aktivitet kan forblive stabil og dermed hjælpe med at omprogrammere den cirkadiske funktion.

 

Så mens det nu er kendt, hvordan NAD+ påvirker den cirkadiske rytme, ønskede forskerne at finde ud af, om dette faktisk er den grundlæggende årsag til NAD+'s velkendte sundhedsmæssige fordele.

 

For at undersøge dette blev to grupper af mus givet NR i to måneder. Den ene gruppe bestod af unge, ti måneder gamle mus med høje NAD+-niveauer, den anden gruppe bestod af ældre, 22 måneder gamle mus med lave NAD+-niveauer. Begge grupper fik NR i seks måneder. Efter disse seks måneder fandt de, at de gamle mus' undertrykte BMAL1-binding, cellulære oscillationer, respirationsrytmer og aktivitetsrytmer blev genoprettet til ungdommelige niveauer, der var sammenlignelige med den yngre kontrolgruppe.
